Ubisoft has released the first concept art for its upcoming Splinter Cell remake and said it's aiming to create 'a top-tier remake'.

—”to deliver new-generation visuals and gameplay, and the dynamic lighting and shadows the series is known for”.“It’s not simply a remaster,” creative director Chris Auty said in the new anniversary video. “It’s being built from the ground up. We get to do all the content from scratch, fresh and ready to go.”

Associate level design director Zavian Porter continued: “Yeah, we’re aiming to create this top-tier remake and push quality as much as possible. This should help us set a good foundation for the franchise going forward.” Auty added: “We’re very early in production. We’re still prototyping. We don’t want to rush anything. We want to make sure that we absolutely nail the game, we actually do everything in the right way, and produce an absolutely stellar-quality experience. And we will be going dark for a little while, so we can focus on making the absolute best game possible.”

Colon tumour cell death causes mTOR dependence by paracrine P2X4 stimulation - NatureColon tumour cell death causes mTOR dependence by paracrine P2X4 stimulation - NatureChemotherapy-induced death of colon cancer cells causes ATP release triggering P2X4 to mediate an mTOR-dependent pro-survival program in neighbouring cancer cells, which renders them sensitive to mTOR inhibition.

Single-cell RNAseq uncovers cellular heterogeneity and provides a signature for pediatric sleep apneaBackground Obstructive Sleep Apnea (OSA) is a highly prevalent disease and a major cause of systemic inflammation leading to neurocognitive, behavioral, metabolic, and cardiovascular dysfunction in children and adults. However, the impact of OSA on the heterogeneity of circulating immune cells remains to be determined. Methods We applied single-cell transcriptomics analysis (scRNA-seq) to identify OSA-induced changes in transcriptional landscape in PBMC composition which uncovered severity-dependent differences in several cell lineages. Furthermore, a machine learning approach was used to combine scRNAs-seq cell-specific markers with those differentially expressed in OSA. Results scRNA-seq demonstrated OSA-induced heterogeneity in cellular composition and enabled the identification of previously undescribed cell types in PBMCs. We identified a molecular signature consisting of 32 genes, which distinguished OSA patients from various controls with high precision (AUC=0.96), and accuracy (93% PPV and 95% NPV, respectively) in an independent PBMC bulk RNA expression dataset. Conclusion OSA deregulates systemic immune function and displays a molecular signature that can be assessed in standard cellular RNA without the need for pre-analytical cell separation, thereby making the assay amenable to application in a molecular diagnostic setting.
