. In brief, macrophage-specific miR-21–deficient mice were generated by crossing miR-21–floxed mice withpromotor that is specifically active in macrophages. All mice were bred in C57BL/6N genetic background. All animal studies were performed in accordance with relevant guidelines and regulations of the responsible authorities and approval was obtained from the IRB at the Regierung von Oberbayern . Mice were housed in rooms maintained at constant temperature and humidity with a 12 h light cycle.

The forced oscillation perturbation Prime-8 was performed. This volume-driven, 8 s long measurement subjects the lung to a standardized test signal of oscillatory frequencies well above and below the subject´s ventilation frequency. Different frequencies will probe different regions of the lung. The measurements were fitted to constant phase model to obtain Newtonian resistance , tissue damping and tissue elastance .

Immune cell-enriched fractions from the lung were either stained with antibodies against CD45-FITC , F4/80-PECy7 , CD11b-PE and CD24-PE/Dazzle 594 to isolate macrophages, or with antibodies against CD45-FITC , Ly6G-PE/Dazzle 594 , CD3-PECy7 and CD19-PECy5 to isolate neutrophils and T cells.
